The observation that neuropeptide Y (NPY) is the most abundant peptide present in the mammalian nervous system and the finding that it elicits the most powerful orexigenic signal have led to active investigations of the properties of the NPY family of hormones, including peptide YY (PYY) and pancreatic polypeptide (PP). Nearly two decades of research have led to the identification of several NPY receptor subtypes and the development of useful receptor selective ligands. Moreover, these investigations have imp- cated NPY in the pathophysiology of a number of diseases, including feeding disorders, seizures, memory loss, anxiety, depression, and heart failure. Vigorous efforts are therefore continuing, not only to understand the bioche- cal aspects of NPY actions, but also toward developing NPY-based treatments for a variety of disorders. To facilitate these efforts, it was decided to produce the first handbook on NPY research techniques as part of the Methods in Molecular Biology Series. In compiling Neuropeptide Y Protocols, I have gathered contributions on techniques considered critical for the advancement of the NPY field from experts in various disciplines. Each chapter starts with a brief introduction, with Materials and Methods sections following. The latter sections are presented in an easy to follow step-by-step format. The last section of the chapter, Notes, highlights pitfalls and the maneuvers employed to overcome them. This information, not usually disseminated in standard research pub- cations, may prove extremely useful for investigators employing these te- niques in NYP research.

“Have you tried peptides? Small proteins, the best in the land! Won’t you try peptides? Keep all your body processes in hand! For labor and lactation oxytocin you must buy! Enkephalin always gives a good runner’s high! So won’t you try peptides? Small proteins, the best in the land!” The above words [1], penned by Gary Gisselman to open Peptide Ångst: La Triviata, the opera which made its world premiere on July 1, 1999, also serve as a fitting charge to the th 16 American Peptide Symposium. This latest edition of a premier biennial series was held under the auspices of the American Peptide Society, June 26–July 1, 1999, at the Minneapolis Convention Center, Minneapolis,Minnesota, with the undersigned serving as Co-Chairs. The fortunate coincidence of the calendar allowed us to set as the theme “Peptides for the New Millennium”, and in our judgment, the approximately 1200 participants [2] who converged in the Twin Cities from academic and industrial institutions in 36 countries were treated to an exciting and stimulating conference that left most everyone with an enthusiastic vision for the future of our field. The present Proceedings volume should serve as a handy reference source and succinct snapshot of peptide science at essentially its century mark – the clock having started with the initial contributions of Emil Fischer and Th. Curtius.

The purpose of the preface is to explain the book's objectives and how to use it; give warnings, disclaimers, and the like.* The main objective of Protein and Peptide Analysis by Mass Spec trometry is quite straightforward―to present authoritative, up-to-date, and practical accounts of the use of mass spectrometry in the analysis of pep tides and proteins. How to use it? Every reader will have their own particular interests and will surely be drawn toward the chapters that cover these interests. Within the remaining chapters, however, techniques are described with analytical possibilities that such a reader can then only guess at. So, read the book fully. Again, as is customary in the Methods in Molecular Biology series, the chapter format (Introduction, Materials, Methods, and Notes) allows the authors to introduce the techniques, to explain their relevance and applicability, and, above all, to provide detail―detail that represents each author's accumulated experience and enables the reader to use and benefit from these methods. So, read the book fully, and read it diligently. Warnings and disclaimers: Mass spectrometry today offers the pro tein chemist ready access to a wealth of information that is otherwise avail able only with great difficulty, or perhaps not at all. With this goal in sight, any warnings and disclaimers will almost surely be ignored. So, a warning anyway; the use of mass spectrometry might be habit forming.
